I am currently the second most senior member of the Department and have the most experience in molecular biology, and I would like to continue as principal investigator of this award. My research group has been involved in the molecular, cellular and developmental biology of calico cats and their mutant relatives. My particular interest has been the structure and function of the genome of the calico cat and its gene products. We have previously shown that the trans-dominant mutations responsible for the absence of cutaneous melanin in the calico cat also affect the synthesis of tyrosinase. We have made the clinical observation that this gene defect also alters the natural skin color of the white cat from bright red to beige. A variety of animal models have been generated to study the biochemistry, cell biology, and physiology of melanin and tyrosinase. We have shown that differences in tyrosinase protein may account for variations in pigmentation. We have made additional observations on the mutations in the calico gene that affect both the synthesis and the activity of tyrosinase. Our principal interest is how the tyrosinase gene and its protein are regulated and if the differences between the normal and the mutated protein are significant in terms of function. We have shown that the transcription of the tyrosinase gene in the melanocyte is regulated by both cell type and cell state. We are interested in the upstream control mechanisms that effect tyrosinase gene expression. We have found that the melanocyte is very sensitive to cell density. If the cells are separated into multiple layers, they will not divide and the tyrosinase gene will remain inactive.
